Abstract

A recent review of one of my grant applications commented on the ‘rediscovery of tropomyosin’. I was tempted to write back in my rebuttal to the reviewer that I didn’t realise it had been lost. Uncharacteristic maturity prevailed and I resisted the temptation, but I was struck by the underlying observation that research on the str- ture and function of tropomyosin has been somewhat invisible, particularly in terms of the cytoskeleton isoforms. So, how can it be that one of the two major components of the actin filament has been so thoroughly overlooked? I suspect that the answer is disappointingly pedestrian. Whereas the biochemistry of the 1980s revealed the potential of tropomyosin isoforms to diversify the function of actin filaments, the subsequent disenchantment with isoform biology in general in the 1990s inhibited growth of this field. With the development of more sophisticated experimental - proaches we are now seeing a growing realisation of the importance of tropomyosin in regulating actin filaments beyond its pivotal role in muscle contraction. The opportunity to edit this book came at a time when we had written several reviews on different aspects of tropomyosin function and I had just finished the background reading for a comprehensive review of tropomyosin biology. I realised that the field was simply beyond the capacity of any one person to do the field j- tice.

Abstract

The epidermal growth factor (EGF) receptor and its downstream signal transduction networks have been implicated in the ontology and maintenance of tumor tissues, which has motivated the discovery and development of molecularly targeted anti-EGFR therapies. Edited by John Haley and William J. Gullick, EGFR Signaling Networks in Cancer Therapy, is separated into two sections. The first of which probes the molecular pathways and the intersection of signaling networks which are frequently deregulated in human cancers, with a view to describing EGF receptor in a tumor tissue specific context. Meanwhile, the second section illustrates the many ways in which EGF receptor contribute to abnormal survival and migration signaling in cancer cells and to epithelial to mesenchymal transition and metastasis. The book also describes the mitogenic, survival, adhesive and migratory pathways within a framework of interacting subsystems that contribute to the activity and physiological regulation of the receptor in normal and neoplastic tissues. Even though there is still much to learn, this volume explores this fascinating system with compelling information.

Abstract

Cyclin Dependent Kinase 5 provides a comprehensive and up-to-date collection of reviews on the discovery, signaling mechanisms and functions of Cdk5, as well as the potential implication of Cdk5 in the treatment of neurodegenerative diseases. Since the identification of this unique member of the Cdk family, Cdk5 has emerged as one of the most important signal transduction mediators in the development, maintenance and fine-tuning of neuronal functions and networking. Further studies have revealed that Cdk5 is also associated with the regulation of neuronal survival during development as well as in neurodegenerative diseases. These observations indicate that precise control of Cdk5 is essential for the regulation of neuronal survival. The pivotal role that Cdk5 appears to play in both the regulation of neuronal survival and synaptic functions thus raises the interesting possibility that Cdk5 inhibitors may have therapeutic potential for the treatment of a number of neurodegenerative diseases.

Abstract

Complement has long been regarded as a pivotal effector arm of the innate immune response, eliciting important immunoregulatory functions in the context of inflammation and also serving as a vital link between the innate and adaptive immune response. In the post-genomic era, our knowledge of the innate immune system is enriched by findings that point to novel functions that do not strictly correlate with immunological defense and surveillance, immune modulation or Inflammation. Several studies indicate that complement proteins exert functions that are either more complex than previously thought, or go well beyond the innate immune character of the system. The advent of high-throughput platforms for genome and proteome-wide profiling, together with the enormous amount of raw genetic information that has accumulated in the databases, have stirred new expectations in biomedical research. They have led complementologists to revisit established biological systems, such as the complement system, from a global and integrative perspective. Complement research is now faced with the challenge of trying to integrate isolated biochemical pathways into complex gene and protein regulatory circuits. In this respect, scientists from around the world convened at the Fourth Aegean Conferences Workshop on Complement Associated Diseases, Animal Models, and Therapeutics (June 10-15, 2007), to discuss recent advances in this fast evolving field. Abstract

A great deal of misinformation is present in popular culture regarding the effects of creatine supplementation on health and sports performance. For instance, it is not uncommon for various media outlets to claim, in spite of the lack of supporting evidence, that 'supplementing with creatine is harmful to the liver and kidneys and may cause dehydration and cramping.' In reality, creatine is intimately involved in energy metabolism, performance, and training adaptations, and, recently, several studies have uncovered its potential clinical application toward treating various neuromuscular diseases and maintaining brain function. This book unifies the sum of information on how creatine affects body composition, exercise performance, and health. Because the body of data on creatine is ample and constantly growing, a book such as this is a timely and important resource for the clinician, the coach, and the sports scientist.

Abstract

Specific complexes of protein and RNA carry out many essential biological functions, including RNA processing, RNA turnover, RNA folding, as well as the translation of genetic information from mRNA into protein sequences. Messenger RNA (mRNA) decay is now emerging as an important control point and a major contributor to gene expression. Continuing identification of the protein factors and cofactors, and mRNA instability elements, responsible for mRNA decay allow researchers to build a comprehensive picture of the highly orchestrated processes involved in mRNA decay and its regulation.
